X射線(xian)衍射分析已成(cheng)為(wei)(wei)當今藥(yao)(yao)(yao)(yao)物(wu)(wu)研究與開(kai)發中普遍應用的(de)(de)(de)(de)(de)一(yi)種(zhong)物(wu)(wu)理分析方法(fa)和常規檢測技(ji)(ji)術(shu)(shu),而且(qie)在(zai)化(hua)學(xue)藥(yao)(yao)(yao)(yao)研究與質量控(kong)制方面正(zheng)發揮著其(qi)(qi)他分析技(ji)(ji)術(shu)(shu)不(bu)可代替的(de)(de)(de)(de)(de)重要(yao)(yao)作用。化(hua)學(xue)藥(yao)(yao)(yao)(yao)物(wu)(wu)的(de)(de)(de)(de)(de)制藥(yao)(yao)(yao)(yao)研究過(guo)程中,粉(fen)末X射線(xian)衍射技(ji)(ji)術(shu)(shu)主要(yao)(yao)應用于固體狀態下單一(yi)化(hua)合(he)(he)物(wu)(wu)的(de)(de)(de)(de)(de)鑒別與晶(jing)(jing)(jing)型(xing)(xing)(xing)確定(ding)(ding)、晶(jing)(jing)(jing)態與非晶(jing)(jing)(jing)態物(wu)(wu)質判斷、多(duo)(duo)種(zhong)化(hua)合(he)(he)物(wu)(wu)組成(cheng)的(de)(de)(de)(de)(de)多(duo)(duo)相體系中的(de)(de)(de)(de)(de)組分分析(定(ding)(ding)性或定(ding)(ding)量)。化(hua)學(xue)結(jie)構(gou)相同(tong)(tong)的(de)(de)(de)(de)(de)藥(yao)(yao)(yao)(yao)物(wu)(wu),可因結(jie)晶(jing)(jing)(jing)條件不(bu)同(tong)(tong)而得到不(bu)同(tong)(tong)的(de)(de)(de)(de)(de)晶(jing)(jing)(jing)型(xing)(xing)(xing),這(zhe)種(zhong)現象稱(cheng)為(wei)(wei)多(duo)(duo)晶(jing)(jing)(jing)型(xing)(xing)(xing)。藥(yao)(yao)(yao)(yao)物(wu)(wu)的(de)(de)(de)(de)(de)晶(jing)(jing)(jing)型(xing)(xing)(xing)不(bu)同(tong)(tong),其(qi)(qi)物(wu)(wu)理性質如密(mi)度、熔點、溶(rong)解(jie)度和溶(rong)出(chu)速(su)度均有(you)不(bu)同(tong)(tong)。在(zai)一(yi)定(ding)(ding)溫度與壓力下,多(duo)(duo)晶(jing)(jing)(jing)型(xing)(xing)(xing)中只有(you)一(yi)種(zhong)是(shi)穩(wen)(wen)定(ding)(ding)型(xing)(xing)(xing),溶(rong)解(jie)度zui小(xiao),化(hua)學(xue)穩(wen)(wen)定(ding)(ding)性好,而其(qi)(qi)他晶(jing)(jing)(jing)型(xing)(xing)(xing)則為(wei)(wei)亞穩(wen)(wen)定(ding)(ding)型(xing)(xing)(xing),它們(men)zui終可轉變為(wei)(wei)穩(wen)(wen)定(ding)(ding)型(xing)(xing)(xing)。但由于從亞穩(wen)(wen)定(ding)(ding)型(xing)(xing)(xing)轉變為(wei)(wei)穩(wen)(wen)定(ding)(ding)型(xing)(xing)(xing)的(de)(de)(de)(de)(de)過(guo)程通(tong)常比較緩慢,因此許(xu)多(duo)(duo)結(jie)晶(jing)(jing)(jing)藥(yao)(yao)(yao)(yao)物(wu)(wu)都存(cun)在(zai)多(duo)(duo)晶(jing)(jing)(jing)現象。亞穩(wen)(wen)定(ding)(ding)型(xing)(xing)(xing)的(de)(de)(de)(de)(de)溶(rong)解(jie)度大,故(gu)溶(rong)出(chu)速(su)度也較快,一(yi)般來講,亞穩(wen)(wen)定(ding)(ding)型(xing)(xing)(xing)的(de)(de)(de)(de)(de)生(sheng)物(wu)(wu)利用度高,為(wei)(wei)有(you)效(xiao)(xiao)晶(jing)(jing)(jing)型(xing)(xing)(xing),而穩(wen)(wen)定(ding)(ding)晶(jing)(jing)(jing)型(xing)(xing)(xing)藥(yao)(yao)(yao)(yao)物(wu)(wu)往往低效(xiao)(xiao)甚至無效(xiao)(xiao)。因此,藥(yao)(yao)(yao)(yao)物(wu)(wu)多(duo)(duo)晶(jing)(jing)(jing)型(xing)(xing)(xing)的(de)(de)(de)(de)(de)研究已經成(cheng)為(wei)(wei)新藥(yao)(yao)(yao)(yao)開(kai)發和審(shen)批,藥(yao)(yao)(yao)(yao)物(wu)(wu)的(de)(de)(de)(de)(de)生(sheng)產和質量控(kong)制以及(ji)新藥(yao)(yao)(yao)(yao)劑(ji)型(xing)(xing)(xing)確定(ding)(ding)前(qian)設計所*的(de)(de)(de)(de)(de)重要(yao)(yao)組成(cheng)部分。
